ARH9 (originally rhoC), a member of the RHO gene subfamily (in turn, belonging to the RAS superfamily), was initially assigned to the telomeric region of Chr 5q by isotopic chromosomal in situ hybridization and Southern analysis of somatic cell hybrid DNAs (Cannnizzaro-1990; Huebner-1990). In those experiments, the gene was noted to cosegregate 100% concordantly with the human CSF1 gene in somatic cell hybrids carrying partial Chr 5, together with other human chromosomes. Since the reassignment of the CSF1 locus from Chr 5q to Chr 1p has recently been performed (Morris-1991; see GEM:01p/CSF1), the same authors also reexamined the localization of the ARH9 gene. New results demonstrated that ARH9 was present in hybrids retaining Chr 1p, but not 5 (Morris-1993). The authors mapped ARH9 relative to other 1p loci, localizing the gene to the region 1p13-p31. Fluorescence in situ hybridization (FISH) to metaphase chromosomes with a 12 kb genomic ARH9 clone confirmed the gene's localization to Chr 1, within bands p21-p13. Explaining the previous erroneous localization of the gene (based on usage of isotopic in situ hybridization), the authors suggested that this error 'emphasizes a significant limitation of this technique, particularly, the low signal-to-noise ratio' in comparing with the FISH method."
