[1] The holoenzyme is presumed to be a homotetramer. It catalyzes the energetically important stage of glycolysis - oxidative phosphorylation.

Benham-1989 confirmed the presence of a single expressed locus for this major glycolytic enzyme on 12p13. One proved pseudogene is mapped to Chr Xp11 (GEM:0Xp11/GAPDP1). Over 15 GAPD-like sequences have been revealed, they being dispersed among various chromosomes, including Chr X: GEM:01^/GAPDL1; GEM:0X^/GAPDL2; GEM:02q/GAPDL3; GEM:04^/GAPDL4; GEM:06^/GAPDL5; GEM:15^/GAPDL6; GEM:08^/GAPDL7; GEM:06^/GAPDL8; GEM:01^/GAPDL9; GEM:0X^/GAPDL10; GEM:18^/GAPDL11; GEM:0Xq/GAPDL12; GEM:07^/GAPDL13; GEM:13q12/GAPDL14; GEM:11^/GAPDL15; GEM:05q3/GAPDL16."
