[1] It is assumed that, in addition to ten CON and J genes, about 100-200 VAR minigenes (both coding and pseudogenes) are located in subsegment 14q32.33, within a 2.5 Mb interval. On one hand, these minigenes have the common evolutionary origing; but on the other hand, they are numerous and have undergone the divergent evolution as to their primary structure. Therefore, the minigenes are subdivided into several homology groups, the intra-group similarity between copies is greater than inter-group one. Six homology classes (I-VI) are distinguished for VH minigenes. The pricise number of coding and faulty (pseudogene) copies in each class is not determined yet, however, the following numeric distribution among classes seems to be credible: class I, at least 25; class II, at least 10; class III, aboit 30; class IV, at least 10 minigenes; class V, three coding genes; and class VI, one coding gene. In this case, it is difficult to elaborate the gene denotation, since it would be reasonable to reflect in the gene symbol both assignment to a homology group and the minigene ordinal number within the group. To meet both conditions, the GEM catalogue adopted the letter denotation for homology groups (A to F). On the other hand, it is hardly reasonable to register each indiviual gene, until there is enough information to individualize each of many tens of them.
[2] VH minigenes belonging to A-E (I-V) homology groups are located within a 579 kb NotI fragment at the telomeric end of the whole IGH cluster, whereas VF1 (or V6) minigene flanks the distal end of the J-DIV minigene cluster, which precedes the bulk of VH genes; see also FAM:IGH/14q3233."
