De Benedetti et al. (1989) isolated a cDNA clone, named K16, by screening a human cDNA library with antibodies directed against human placental hexokinase. The same antibodies inactivate human hexokinase I (which maps to chromosome 10) but do not recognize human hexokinase II. The sequence of the cDNA clone showed a typical kinase domain but was different from the sequence of any other known kinase.
De Benedetti et al. (1989) mapped the genomic sequence corresponding to K16 to the 7q22-q23 region using a human-hamster somatic cell hybrid panel and a hybrid carrying a single copy of human chromosome 7 with an interstitial deletion of 7q22-q32. RFLPs were identified. Since the CF locus maps in the same region, De Benedetti et al. (1989) tested linkage between the RFLPs and the CF locus. The locus may be situated proximal to CF.
