The CD53 antigen was recognized by 4 monoclonal antibodies submitted to the Fourth International Workshop on Human Leucocyte Differentiation Antigens. Hadam (1989) demonstrated that it is the most specific and reliable panleukocyte marker. Angelisova et al. (1990) showed that CD53 is an N-glycosylated panleukocyte antigen with a molecular mass between 35 and 42 kD. From a cDNA clone, they deduced that it contains 219 amino acids and probably consists of 4 transmembrane regions and a major extracellular hydrophilic loop containing 2 potential N-glycosylation sites. Angelisova et al. (1990) suggested that the CD53 glycoprotein is the true homolog of the rat MRC-OX-44 antigen.
Amiot (1990) also isolated a cDNA corresponding to the gene encoding the CD53 antigen. The sequence of the cDNA predicted a protein of 219 residues bearing 4 putative membrane-spanning hydrophobic domains.
By study of human/rodent somatic cell hybrids, Virtaneva et al. (1993) demonstrated that the CD53 gene is located on chromosome 1p31-p12. By fluorescence in situ hybridization, Taguchi et al. (1993) assigned the human gene to 1p21-p13.3 and the rat gene to 2q34-q41. By interspecies backcross mapping, they assigned the gene to the midportion of mouse chromosome 3. By fluorescence in situ hybridization, Gonzalez et al. (1993) mapped the CD53 gene to 1p13.
