Alcalay and Toniolo (1988) obtained a full-length SLC10A3 clone, which they called P3, from placenta and human teratocarcinoma cell cDNA libraries. The deduced protein contains 477 amino acids. Northern blot analysis detected variable expression of a 2.1-kb transcript in all cell lines examined. Southern blot analysis suggested that P3 is conserved from yeast to human.
In their review of the SLC10 family, Hagenbuch and Dawson (2004) stated that P3 has been categorized as a sodium-bile acid cotransporter and named SLC10A3 based on its similarity with SLC10A1 (182396) and SLC10A2 (601295). However, no verification of its transport function had been documented. EST database analysis suggested ubiquitous SLC10A3 expression.
Alcalay and Toniolo (1988) determined that the SLC10A3 gene contains 2 exons and spans about 4 kb. The first exon is noncoding and is contained within a CpG island.
Alcalay and Toniolo (1987) identified 2 expressed genes, P3 and GDX (312070), in the Xq28 band. Their location was established by the discovery of several overlapping lambda and cosmid clones that linked them to the G6PD gene (305900). The relationship of the 2 genes to each other and to G6PD is as follows: G6PD--3-prime--(7 kb)--5-prime--P3--3-prime--(0.5 kb)--5-prime--GDX.
