The 'Polycomb' group (PcG) of genes in Drosophila is required for the maintenance of repression of a number of key developmental regulatory genes. PcG proteins are thought to form a multimeric complex that modifies local chromatin structure and establishes a heritable repression state at particular loci. By carrying out BLAST searches, Coulson et al. (1998) identified a human cDNA, called PHF1, that was similar to the Drosophila 'Polycomblike' (Pcl) gene. They used the cDNA to screen a placenta library and isolated cDNAs containing the complete coding region of PHF1. The predicted 462-amino acid protein contains 2 PHD fingers. Northern blot analysis revealed that PHF1 is expressed as 2.6- and 3-kb transcripts in various tissues.
By fluorescence in situ hybridization, Coulson et al. (1998) mapped the PHF1 gene to 6p21.3.
