Meacci et al. cloned a cDNA for myocardial cGMP-inhibited cAMP phosphodiesterase from a human heart cDNA library. They obtained direct proof that the cAMP hydrolytic activity was inhibited by cGMP (Meacci-1992)."

GEN

The open reading frame encoded a protein of 125 kD. A single mRNA species hybridized with a 4-kb EcoRI restriction fragment of the cDNA containing the entire open reading frame. The carboxyl-terminal region of the deduced amino acid sequence contained a putative catalytic domain conserved among mammalian PDE families (Meacci-1992)."

The cyclic nucleotide phosphodiesterases (FAM:PDE/00.0) regulate the cellular concentrations of cyclic nucleotides and thereby play a role in signal transduction."

The PDE3 family is characterized by high affinity for cAMP, inhibition by cGMP, sensitivity to specific inhibitors such as cilostamide and milrinone, and by phosphorylation and activation in response to insulin and agents that increase cAMP. In human genome another member of PDE3 family is mapped to Che 11p15 (GEM:11p15/PDE3B)."
