GNAI3 gene consists of 8 exons and 7 introns. The structure is identical to the gene for alpha-I2 peptide (GEM:03p21/GNAI2). The human mRNA is 2.6 kb in size. The promoter contains a TATA-like box, TTTAAA being located 182 bp before the start codon, and the GC blocks being located 87, 300, and 400 bp before the start codon. Typical CCAAT blocks are lacking."

The amino acid sequence of human alpha-I3 protein differs from that of bovine alpha-I protein by 6% and from rat alpha-I2 protein, by 15%. The coding regions of the human gene and rat GI3S gene exhibit a pronounced similarity (92%). The homology of their coding regions is 80%."
