Transcriptional regulation is exerted through protein factors that bind to distinct DNA elements in the vicinity of target genes. Many control elements are recognized by families of transcription factors, opening combinatorial possibilities to the regulatory interactions. Among the most widely distributed control elements are GC boxes and related motifs bound by Sp1."

There are several genes encoding Sp transcription factors in human genome: GEM:00.0/SP2; GEM:02q31/SP3; GEM:07p15/SP4. The Kalff-Suske et al. noted the striking colocalization of each of the Sp factors with the homeo box gene clusters HOX A (FAM:HOXA/07p1), HOX C (FAM:HOXC/12q1), and HOX D (FAM:HOXD/02q31) on the corresponding chromosomes (Kalff-Suske-1996)."
