The locus have been mapped on the basis of the close linkage with Rhesus antigen in families with uni- or bilateral breast cancer and/or with primarilly multiple (uni- or bilateral) ovarian cancer. In some cases, the relatives with cystic fibrous degeneration of mammary glands (but not fibradenoma) were also included in the counting of the general prevalence (i.e., in the number of affected with incomplete gene expression) to increase the informativeness."

Since several genes that are directly or indirectly involved in carcinogenesis are mapped in this region of chr 1p3 (GEM:01p36/A12M2, GEM:01p36/CMM, GEM:01p3/ERVPL1, GEM:01p36/FGR, GEM:01p32/BLYM, GEM:01p32/MYCL1, GEM:01p3/JUN, GEM:01p3/LCK, GEM:01p36/NBL1, and GEM:01p3/RAB3B), the hypothesis of independence of the BCS2 locus requires additional verification."
